Junshi Biosciences Announces Submission of a Marketing Authorization Application for Toripalimab to the UK Medicines and Healthcare Products Regulatory Agency

— 2nd MAA submission in Europe reflects toripalimab’s steady yet speedy progress towards global commercialization

SHANGHAI, China, Nov. 24, 2022 (GLOBE NEWSWIRE) — Shanghai Junshi Biosciences Co., Ltd (“Junshi Biosciences”, HKEX: 1877; SSE: 688180), a leading innovation-driven biopharmaceutical company dedicated to the discovery, development, and commercialization of novel therapies, announced today that the company has submitted a marketing authorization application (“MAA”) to the United Kingdom’s Medicines and Healthcare products Regulatory Agency (“MHRA”) for toripalimab. The indications requested in the MAA are: 1) Toripalimab, in combination with cisplatin and gemcitabine, for the first-line treatment of patients with locally recurrent or metastatic nasopharyngeal carcinoma (“NPC”) and 2)Toripalimab, in combination with paclitaxel and cisplatin, for the first-line treatment of patients with unresectable locally advanced/recurrent or metastatic esophageal squamous cell carcinoma (“ESCC”). Junshi Biosciences also submitted a MAA to the European Medicines Agency (“EMA”) for toripalimab for the same indications in mid-November.

The MAA submission for NPC and ESCC is based on the results from JUPITER-02 (an international randomized, placebo-controlled, double-blinded Phase III trial, NCT03581786) and JUPITER-06 (a randomized, placebo-controlled, double-blinded, multi-center Phase III trial, NCT03829969).

The JUPITER-02 results were first presented in the plenary session of the American Society of Clinical Oncology (“ASCO”) 2021 annual meeting (#LBA2) and subsequently published in detail as the cover article of the September 2021 issue of Nature Medicine. According to its final progression-free survival (PFS) analysis, the toripalimab plus chemotherapy arm had a significantly longer PFS than the placebo plus chemotherapy arm as assessed by a blinded independent radiology committee (BIRC) with median PFS of 21.4 vs. 8.2 months, hazard ratio (HR)=0.52 (95% CI: 0.37-0.73), two-sided p<0.0001.

In 2021, two indications for the treatment of NPC were approved by the China National Medical Products Administration (“NMPA”), thereby making toripalimab the world’s first immune checkpoint inhibitor approved for the treatment of NPC. In the the United States, the NPC indications of toripalimab have been granted 2 Breathrough Therapy Designations and 1 Orphan Drug Designation by the Food and Drug Administration (“FDA”), while the Biologics License Application (“BLA”) for toripalimab for the treatment of NPC is under review. If approved, toripalimab will be the first and only drug approved for the treatment of NPC in the U.S. In Europe, toripalimab was designated as an orphan medicinal product by the European Commission (“EC”) for the treatment of NPC.

The JUPITER-06 results were first presented in a mini-oral session during the 2021 European Society for Medical Oncology (“ESMO”) Congress and later published in Cancer Cell with an editorial preview. The overall survival (OS) and PFS were significantly better in the toripalimab plus chemotherapy arm than in the placebo plus chemotherapy arm, with a median OS of 17 vs. 11 months (HR=0.58, 95% CI: 0.43-0.78, P=0.0004) and PFS HR=0.58 (95% CI: 0.46-0.74), p<0.0001.

In May 2022, the supplemental new drug application (“sNDA”) for toripalimab in combination with paclitaxel and cisplatin for the first-line treatment of patients with unresectable locally advanced/recurrent or distant metastatic ESCC was approved by the NMPA in China. Additionally, US FDA has also granted an orphan drug designation to toripalimab for the treatment of patients with ESCC.

About Nasopharyngeal Carcinoma
NPC is a malignant tumor that occurs in the nasopharyngeal mucosal epithelium and is one of the most common head and neck cancers in China. According to the World Health Organization, the number of newly diagnosed NPC cases in 2020 has exceeded 130,000 worldwide. Due to the location of the primary tumor, surgery is rarely an option, and patients with localized disease are treated primarily with chemotherapy and radiotherapy. In the United States and Europe, there are presently no drugs approved for the treatment of NPC, for which recommendations for initial treatment (gemcitabine and cisplatin) are based on randomized trials conducted in China.

Esophageal cancer is one of the most common malignant tumors in alimentary tract. According to data released by GLOBOCAN 2020, 604,100 new esophageal cancer cases and 544,076 deaths due to esophageal cancer occurred globally. The incidence and death rates of esophageal cancer ranked seventh and sixth among all malignant tumors around the world. ESCC and esophageal adenocarcinoma are the two main histological subtypes of esophageal cancer. For patients with advanced ESCC, recently updated ESMO guidelines recommend a platinum-fluoropyrimidine doublet with a PD-1 blocking antibody for treatment of locally advanced or metastatic ESCC. Of note, the indications for those PD-1 inhibitors approved in Europe are restricted to a subset of patients with ESCC. Therefore, there is an urgent unmet need for new drugs and treatments to extend the survival of patients with ESCC, particularly those with low PD-1 tumor expression.

Toripalimab is an anti-PD-1 monoclonal antibody developed for its ability to block PD-1 interactions with its ligands, PD-L1 and PD-L2, and for enhanced receptor internalization (endocytosis function). Blocking PD-1 interactions with PD-L1 and PD-L2 promotes the immune system’s ability to attack and kill tumor cells.

More than thirty company-sponsored toripalimab clinical studies covering more than fifteen indications have been conducted globally by Junshi Biosciences, including in China, the United States, Southeast Asia, and European countries.   5. in combination with paclitaxel and cisplatin in first-line treatment of patients with unresectable locally advanced/recurrent or distant metastatic ESCC;
  6. in combination with pemetrexed and platinum as the first-line treatment in EGFR mutation-negative and ALK mutation-negative, unresectable, locally advanced or metastatic non-squamous non-small cell lung cancer (“NSCLC”).

The first three indications have been included in the National Reimbursement Drug List (“NRDL”) (2021 Edition). Toripalimab is the only anti-PD-1 monoclonal antibody included in the NRDL for treatment of melanoma and NPC.

In the United States, the FDA is reviewing the BLA resubmission for toripalimab in combination with gemcitabine and cisplatin as first-line treatment for patients with advanced recurrent or metastatic NPC and for toripalimab monotherapy for the second-line or later treatment of recurrent or metastatic NPC after platinum-containing chemotherapy. The FDA has set a PDUFA action date for December 23, 2022. The FDA has granted Breakthrough Therapy designations for toripalimab in combination with chemotherapy for the first-line treatment of recurrent or metastatic NPC as well as for toripalimab monotherapy in the second or third-line treatment of recurrent or metastatic NPC. Additionally, the FDA has granted Fast Track designation for toripalimab for the treatment of mucosal melanoma and Orphan Drug designations for the treatment of esophageal cancer, NPC, mucosal melanoma, soft tissue sarcoma, and small cell lung cancer (“SCLC”).

East Asia intergovernmental congress expected to forge cooperation

West Java Governor Ridwan Kamil has said that he expects the 11th East Asia Local and Regional Government Congress in Bandung City to strengthen cooperation in several fields.

The congress is being held from November 23–25, 2022.

“Although this event is being held amid a state of mourning for the Cianjur earthquake (victims), I hope this activity will result in mutually strengthening cooperation in various fields,” he said while opening the congress here on Thursday.

He expressed the hope that the congress will result in real cooperation for the progress of Asian nations.

“I also hope that there will be a blueprint for dealing with the economy after the COVID-19 pandemic,” he said.

It is also hoped that all delegates will feel comfortable in Bandung, and the congress can become the best momentum for determining the future of East Asia, the governor added.

“Welcome, East Asian mayors/district heads and governors in Bandung city,” he remarked.

Kamil said he believes that the successful implementation of the recent G20 Summit in Bali ignited West Java’s spirit to host the congress well.

“There are many obstacles, but I hope that there is cooperation that can strengthen each other. This congress is very strategic in discussing many things about lots of knowledge,” he added.

Meanwhile, the Japanese Ambassador to Indonesia, Kenji Kanasugi, said that West Java and Japan have enjoyed a long relationship. Therefore, he expressed the hope that there will be cooperation in the tourism sector, which has been affected by the COVID-19 pandemic.

“(There should be cooperation in) the promotion of tourism that was affected by the COVID-19 pandemic. I hope that all the delegates will help increase development in many ways,” he added.

Meanwhile, the Governor of Nara Prefecture, Japan, Arai Shogo, called on all delegates to work together to create a better future for East Asia.

“In this congress, the focus is on tourism and economic development after the COVID-19 pandemic,” he said.

West Java and Shizuoka Prefecture will cooperate in the field of human resources and economic development, he added.

No increase in acute kidney injury cases: ministry

The number of cases of atypical progressive acute kidney injury in children has not increased in the past two weeks, Health Ministry spokesperson Mohammad Syahril informed on Thursday.

“Since two weeks ago until now, there have been no additional cases. This is the result of a joint effort, where there have been no additional cases and no deaths,” Syahril said at a webinar on the management of acute kidney injury, which was followed from Jakarta.

Currently, there are 11 patients undergoing treatment for acute kidney failure at the National Central General Hospital Dr. Cipto Mangunkusumo (RSCM) Jakarta, he added.

“We hope that the 11 patients who are still being treated at RSCM can recover after being given the Fomepizole antidote that we have brought in,” he said.

The Ministry of Health recorded a total of 324 cases of acute kidney injury in 27 provinces, Syahril informed. The number of cases has remained unchanged since November 2, 2022, he said.

The ministry, in collaboration with the Food and Drug Supervisory Agency (BPOM), examined the alleged link between ethylene glycol (EG) and diethylene glycol (DEG) contamination in syrup drugs and kidney failure, he added.

Health Minister Budi Gunadi Sadikin earlier said that cases of acute kidney injury in Indonesia dropped dramatically after the government temporarily stopped the distribution and use of syrup drugs.

In addition, the Ministry of Health tested the use of Fomepizole on 10 acute kidney injury patients at RSCM Jakarta and the drug was found effective in restoring their health.

After the number of cases of acute kidney injury declined, the government announced a list of safe-to-use syrup drugs based on safety and product quality testing conducted by BPOM on October 23.

2.7 mln MSMEs holding business identification numbers: minister

Currently, 2.7 million micro, small, and medium enterprises (MSMEs) are holding business identification numbers (NIBs), Cooperatives and Small and Medium Enterprises (SMEs) Minister Teten Masduki has said.

“Our target this year was 2.5 million MSMEs to get NIBs. Today, it (the target) has been surpassed. Now, 2.7 million have obtained NIBs. The biggest (group) is micro-enterprises, 2.6 million,” he informed at the Micro-Business Formal Transformation Guard (Garda Transfumi) Appreciation Day event here on Thursday.

According to him, the number of MSMEs in Indonesia has reached 64.2 million, but only half of them need NIBs.

In addition, based on data from the Halal Certification Agency (BPJPH), the estimated number of MSMEs that require halal certification is also around 30 million.

He stressed that the target of halal certification and NIB issuance for 3 million MSMEs per year is too low because it would take ten years to reach around 30 million. Therefore, halal certification and NIBs issuance are being accelerated, he said.

“Next year, we will try to rev again, and maybe later, the work method will be enhanced,” he added.

The minister informed that as many as 10 million MSMEs have been identified as permanent businesses. Thus, those MSMEs would be prioritized for NIBs, halal certificates, and distribution permits, he added.

He further said that data on the 10 million MSMEs would be submitted to the cooperatives and SMEs offices in regions and Garda Transfumi volunteers so that they can get mentorship and obtain NIBs and business certifications.

Garda Transfumi is a collaborative program between the Ministry of Cooperatives and SMEs, Mercy Corp Indonesia, MicroMentor, the MSME Joint Movement (Geber) Association, and other associations that recruit volunteers for Garda Transfumi.

Volunteers of the program assist MSME players in obtaining business permits, in this case, NIBs. Through the assistance, the government is providing protection and convenience to micro-entrepreneurs by helping them legalize their business.

Montara oil spill compensation must benefit people, ecosystem: govt

Coordinating Minister for Maritime Affairs and Investment, Luhut Binsar Pandjaitan, has asked that the compensation for the Montara oil spill be managed professionally for the benefit of the affected communities and ecosystem.

“I also suggest to establish a fishermen’s cooperative to manage it (the compensation) professionally. Later, we will provide assistance, thus the money will not be misspent,” he explained at a press conference on the “Update on the Montara Case Status and Dissemination on Negotiation Results” here on Thursday.

The Montara oil spill occurred from August 21 to November 3, 2009. The oil field was located in the Timor Sea, off the northern coast of Western Australia, Australia.

Given the geographical proximity to Timor Island, the local marine ecosystem, fishermen, and seaweed farmers in East Nusa Tenggara (NTT) province of Indonesia were also affected by the oil spill.

The coordinating minister said that his ministry will promptly assign professionals to calculate the initial capital needed to establish the cooperative so that local seaweed farmers and fishermen can have productive businesses.

“Hence, the people will really become prosperous,” he added.

In addition to improving the people’s welfare, the fund must also be utilized for restoring the maritime environment, which has been damaged by the oil spill, he emphasized.

The attempt is being undertaken to show the world that Indonesia is very serious about tackling environmental pollution, he added.

Thailand-based PTT Exploration and Production (PTTEP), which was operating the oil field at the time of the incident, has agreed to pay a compensation of 192.5 million Australian dollars (equivalent to US$129 million) to fishermen and seaweed farmers in NTT province.

Head of the task force for handling the Montara oil spill case, Purbaya Yudhi Sadewa, informed that the compensation would be disbursed to around 15 thousand fishermen and seaweed farmers.

“Hence, each fisherman (and seaweed farmer) can get AU$6 thousand to AU$7 thousand, more or less,” he noted.

He said that the sum might increase since the task force is still negotiating on the matter.

Although not all parties are satisfied with the compensation, still, it is better than nothing at all, Sadewa added.

The task force is waiting for the court’s decision regarding when the compensation can be distributed.

The government will ensure that the fishermen and seaweed farmers get their rights, he emphasized.

Head of the West Timor Care Foundation, a local non-profit organization, Ferdi Tanoni, said that his organization has accepted the quantum of the compensation.

He said he expected the compensation to be managed professionally for meeting the needs of the local people.
